dc.description.abstract | Cílem této práce je obecné zhodnocení porovnatelnosti metod analýz biofilmu. Teoretická část práce řeší základní pojmy v oblasti biotechnologie vod a základní principy a postupy jednotlivých metod vyhodnocení biofilmu.V dalších částech práce jsou podrobně rozebrány jednotlivé analýzy a jejich výsledky. Odborná část se zabývá provozem biologického reaktoru, kde byly umístěny vlákenné nosiče sloužící pro uchycení biomasy. Důležitým procesem pro provoz reaktoru a tvorbu biofilmu byla nitrifikace. Z nosičů byly v pravidelných intervalech během šesti týdnů odebírány vzorky biofilmu a analyzovány pomocí metod molekulární biologie, respirometrie a optické mikroskopie. Metody molekulárně biologické zahrnují izolaci DNA a následnou kvantitativní polymerázovou řetězovou reakci a FISH analýzu. | cs |
dc.description.abstract | The aim of this work is a general assessment of comparability of biofilm evaluation methods. The theoretical part deals with the basic concepts of water biotechnology and the basic principles and procedures of individual methods of biofilm evaluation.In the following parts of the thesis, the individual analyzes, and their results are analyzed in detail. The essence of the expert part is the solution of the operation of the biological reactor, where the fibrous carriers used for biomass attachment were placed. Nitrification was an important process for reactor operation and biofilm formation. Samples were taken from the carriers at regular intervals over six weeks and analyzed by molecular biology, respirometry and optical microscopy methods. Molecular biological methods include DNA isolation and subsequent quantitative polymerase chain reaction and FISH analysis. | en |
