Abstract
Tato bakalářská práce se zabývá metodou DNA origami. V rešeršní části práce jsou popsány různé přístupy přípravy a analýzy nanostruktur na bázi DNA. Rovněž jsou zmíněny hlavní oblasti využití těchto nanostruktur. Hlavním výsledkem experimentální části je optimalizace přípravy a depozice 2D DNA origami nanostruktur ve tvaru trojúhelníku se stranou cca 120 nm a její modifikace zlatými nanočásticemi (cca 10 nm) pro další experimenty. Kromě optimalizace přípravy nanostruktur byly také provedeny vlastní experimenty studující poškození těchto nanostruktur ionizujícím zářením a první in-singulo experimenty studující radiační poškození DNA fragmentů na těchto nanostrukturách.
The present bachelor's thesis is focused on the method of DNA origami. The literature survey part of the work overviews the main approaches to self-assembled DNA nanostructures and describes the common methods of their visualization and analysis. The main uses of these nanostructures in practice are also mentioned. The main result of the experimental part of the present thesis is optimization of preparation procedures of DNA origami for further experiments with ionizing radiation. Triangular DNA origami were prepared witha side length of cca 120 nm and their variants modified by gold nanoparticles with a diameter of cca 10 nm. In addition to the optimization, further individual research were made into the damage of these nanostructures made by ionizing radiation and first in-singulo experiments studying radiation damage of DNA fragments on these nanostructures.
