Posouzení technik pro hodnocení interakce magnetických nanočástic v biologickém prostředí

Abstract
Tato bakalářská práce se zabývá hodnocením charakteristických vlastností a parametrů nanočástic feromagnetické Fe3O4, nemagnetické Nd2O3 a diamagnetické Zn. Tyto nanočástice mohou být pro biomedicínské aplikace velice užitečné díky svým charakteristickým vlastnostem (např. jako kontrastní činidla při zobrazování magnetickou rezonancí, magnetické označování buněk apod.). Teoretická část čtenáře seznamuje se základními vlastnostmi nanočástic (velikost, tvar apod.) a s metodami, které zkoumají jejich vlastnosti (mikroskopie, toxicita). Výzkumná část se zabývá experimentálním hodnocením toxických účinků nanočástic na dva vybrané bakteriální kmeny, a to Escherichia Coli a Micrococcus Luteus. Byly provedeny testy ekotoxicity a respirometrie, ale i testy cytotoxicity (účinek na jaterní karcinom HepG2). Práce se věnuje především technikám a metodám studia a charakterizaci parametrů a vlastností nanočástic. Výsledkem práce je komparativní srovnání chování buněk, které se dostaly do kontaktu s nanočásticemi a návrh na modifikaci nebo aktualizaci těchto metod pro úspěšné studium s ohledem na jejich (ne)magnetické vlastnosti.
This bachelor thesis deals with the evaluation of characteristic properties and parameters of ferromagnetic Fe3O4, non-magnetic Nd2O3 and diamagnetic Zn nanoparticles. These nanoparticles can be very useful for biomedical applications due to their characteristic properties (e.g. as contrast agents in magnetic resonance imaging, magnetic cell labeling, etc.). The theoretical part acquaints readers with the basic feature of nanoparticles (size, shape, etc.) and with methods that examine their properties (microscopy) and behavior (toxicity). The research part deals with the experimental evaluation of the toxic effects of nanoparticles on two selected bacterial strains; Escherichia Coli and Micrococcus Luteus, where tests of ecotoxicity and respirometry were done. Cytotoxicity tests were performed on hepatic carcinoma HepG2. In particular, techniques and methods for studying the properties of nanoparticles were the subject of this work. The result of the work is a set of comparison of the cell behavior that have come into contact with nanoparticles; also several suggestions of these methods (modification or actualization) were submitted for nanoparticle study with respect to their (non)magnetic properties.
