- ItemA comparative study between chemically modified and copper nanoparticle immobilized Nylon 6 films to explore their efficiency in fighting against two types of pathogenic bacteria(2020-01-05) Swar, Sumita; Máková, Veronika; Horáková, Jana; Kejzlar, Pavel; Parma, Petr; Stibor, IvanHospital-acquired infections (HAI) caused by pathogenic bacteria such as Pseudomonas aeruginosa and Staphylococcus aureus adversely affect public health due to their colonization and subsequent formation of a biofilm on medical devices. Various functionalized polymers including polyamides are commonly used in the biomedical industry aiming to resist bacterial infection as well as to ensure cytocompatibility of the used materials. The present study is focused on the extension of our previous work that developed a new approach to immobilize mPEG on an amine-activated Nylon 6 surface. Herein, we compare functionalized samples of Nylon 6 together with samples additionally containing copper nanoparticles (Cu NPs, size 40-60 nm) physisorbed on a modified surface. Modified samples were analyzed via different techniques including water contact angle measurements, atomic force microscopy and scanning electron microscopy associated with energy dispersive Xray spectroscopy. The copper functionalized samples showed high antibacterial efficacy against Gram-positive Staphylococcus aureus and Gram-negative Pseudomonas aeruginosa. Moreover, most of the prepared samples were cytocompatible.
- ItemDEVELOPMENT OF NANOFIBROUS MATERIALS FOR MEDICAL APPLICATIONS(2022) Horáková, JanaThis habilitation thesis takes the form of comments on the published articles listed at the end of the manuscript. The thesis provides a detailed explanation of my contribution to the published results, since all the articles were written by several coauthors. I selected 9 articles concerning which I am listed as the first or the second author. The articles are dedicated primarily to the electrospinning of biodegradable polymers for the development of vascular grafts, wound dressings and other tissue engineering scaffolds. Thus, the text provides an explanation of specific parts of the research to which I contributed to the field of textile technology and materials engineering. The main part of the thesis is divided into 4 subchapters, the first of which is dedicated to the technology of the fabrication of tubular samples, and provides descriptions of 3 types of electrospinning devices. The second subchapter focuses on the polymers that were used in the attached articles. The third part of the main text is dedicated to methods for the characterization of nanofibrous materials, namely the evaluation of the fibrous morphology and the in vitro assessment of the resulting materials. The final subchapter concerns the modification of electrospun materials. In addition to the summary of the published results of materials modified via the above approaches, the text presents selected data on the investigation of the heparinization of vascular grafts.

It is often a reason for reoperation together with intestinal passage blockage due to formation of peritoneal adhesions. Different materials as local prevention of these complications have been studied, none of which are nowadays routinely used in clinical practice. Nanofabrics created proved to promote healing with their structure similar to extracellular matrix. We decided to study their impact on anastomotic healing and formation of peritoneal adhesions. We performed an experiment on 24 piglets. We constructed 3 hand sutured end-to-end anastomoses on the small intestine of each pig. We covered the anastomoses with a sheet of polycaprolactone nanomaterial in the first experimental group, with a sheet of copolymer of polylactic acid with polycaprolactone in the second one and no fortifying material was used in the Control group. The animals were sacrificed after 3 weeks of observation. Clinical, biochemical and macroscopic signs of anastomotic leakage or intestinal obstruction were monitored, the quality of the scar tissue was assessed histologically, and a newly developed scoring system was employed to evaluate the presence of adhesions. The material is easy to manipulate with. There was no mortality or major morbidity in our groups. No statistical difference was found inbetween the groups in the matter of level of peritoneal adhesions or the quality of the anastomoses. We created a new adhesion scoring system. The material appears to be safe however needs to be studied further to prove itsʹ positive effects.

However, the main prerequisite for the use of such materials in clinical practice consists of the selection of the appropriate sterilization technique. This study is devoted to the study of the impact of traditional sterilization and disinfection methods on a nanofibrous polycaprolactone layer constructed by means of the needleless electrospinning technique. It was determined that hydrogen peroxide plasma treatment led to the loss of fibrous morphology and the creation of a foil. However, certain sterilization (ethylene oxide, gamma irradiation, and peracetic acid) and disinfection techniques (ethanol and UV irradiation) were found not to lead to a change in morphology; thus, the study investigates their impact on thermal properties, molecular weight, and interactions with a fibroblast cell line. It was determined that the surface properties that guide cell adhesion and proliferation were affected more than the bulk properties. The highest proliferation rate of fibroblasts seeded on nanofibrous scaffolds was observed with respect to gamma-irradiated polycaprolactone, while the lowest proliferation rate was observed following ethylene oxide sterilization.

- ItemNanofibrous vascular grafts(Technická Univerzita v Liberci, ) Horáková, JanaV současnosti není v klinické praxi cévní náhrada s vnitřním průměrem pod 6 mm, která by spolehlivě fungovala v dlouhodobém horizontu. Disertační práce se zabývá přípravou maloprůměrových cévních náhrad z biodegradabilních polymerů, které jsou testovány jako potenciálně vhodné materiály pro přípravu tkáňových nosičů pro vaskulární cévní systém. Hlavní myšlenkou tkáňového inženýrství je napodobování přirozeného prostředí - mezibuněčné hmoty. Proto byla ideální cévní náhrada navržena jako dvouvrstvá tubulární struktura s definovanou morfologií vláken. Tato definovaná struktura byla vytvořena elektrostatickým zvlákňováním polykaprolaktonu (PCL). Podobnost morfologie vláken s mezibuněčnou hmotou předpokládá, že po implantaci do organismu proběhne regenerace funkční tkáně. Kromě polymeru polykapronu byl testován polymer ze stejné třídy polyesterů - kopolymer polylatidu a polykaprolaktonu (PLC 70/30). Cévní náhrada připravená z toho polymeru byla tvořena pouze jednou vrstvou.Pro porovnání vlastností polymerů byla provedena charakterizace obou elektrostaticky zvlákněných materiálů. Kopolymer PLC je mírně hydrofilnějšínež polykaprolakton. Termické vlastnosti obou polymerů se značně liší. Zatímco kopolymer PLC je převážně amorfní s teplotou tání okolo 110°C, polykaprolakton je semikrystalický polymer s teplotou tání kolem 57°C. Mechanická pevnost a prodloužení je přibližně desetkrát větší u elektrostaticky zvlákněného kopolymeru PLC než u polykaprolaktonu.Biologické testování elektrostaticky zvlákněných materiálů potvrdilo biokompatibilitu obou testovaných polymerů s fibroblasty i s endotelovými buňkami. Vyšší proliferační stupeň byl pozorován při kultivaci buněk na mírně hydrofilnějším kopolymeru PLC, který zřejmě umožňuje lepší buněčnou adhezi. Vlákenné materiály byly rovněž testovány po interakci s krevními destičkami, které se po inkubaci aktivovaly a agregovaly. Mírnější aktivace byla pozorována po interakci s hladkými foliemi vyrobenými ze stejných materiálů, což dokládá, že na aktivaci destiček má vliv morfologie povrchu. Zvýšená aktivace trombocytů byla naopak pozorována při dynamické inkubaci vlákenných tubulárních vzorků.Vlákenné tkáňové nosiče byly využity jako systém cíleného uvolňování léčiv, konkrétně oxidu dusnatého (NO), který má pozitivní účinky na ardiovaskulární systém. Vlákna polykaprolaktonu byla obohacena o donory NO ze skupiny S-Nitrosothiolů, které umožňují uvolňování NO ve fyziologickém rozmezí po 42 dní v in vitro podmínkách. Po implantaci cévních náhrad jako náhrada břišní části aorty u potkanů bylo zjištěno, že NO inhibuje buněčnou infiltraci do vnitřní a střední vrstvy cévní náhrady. Tento snížený výskyt zánětlivých buněk může bránit vzniku neointimální hyperplazie způsobenou hladkosvalovými buňkami v pozdějších stadiích implantace.

The materials consist of hydrophilic poly(vinyl alcohol) fibers with low (PVA_L) or high (PVA_H) degree of hydrolysis, and hydrophobic poly(ε-caprolactone) (PCL) fibrous layer. Although the PVA_L fibers were fully dissolved following a water exposure, the shape of the scaffold was maintained due to water stable PCL layer. Exposing PVA_H based fibrous layer to water created a hydrogel-like structure with shape defined by the PCL layer. According to the MTT assay, the mouse fibroblasts seeded on the scaffold exhibited the greatest proliferative activity on the PCL fibers. These double-layered scaffolds with different features on each side are very promising for many novel medical applications such as wound dressing or abdominal adhesion prevention.

- ItemVliv hydrofilních/hydrofobních úprav nanovlákenných tkáňových nosičů na buněčnou adhezi(Technická Univerzita v Liberci, 2014) Sykáčková, Pavla; Horáková, JanaRešerše diplomové práce se zabývá problematikou tkáňového inženýrství a biologie obecně. Je zde popsána metoda elektrostatického zvlákňování, parametry ovlivňující tento proces a v neposlední řadě vlastnosti vláknotvorných polymerů. Poslední kapitola rešerše se zabývá tématem smáčivosti, která je v této práci základním kamenem pro vyhodnocování testovaných polymerních materiálů. Polymery byly vybrány k testování pro jejich speciální vlastnosti, jako je biokompatibilita, případně biodegradabilita. Cílem práce bylo elektrostaticky zvláknit vybrané materiály a následně je otestovat. Vyrobené scaffoldy byly podrobeny zkoušce měření kontaktního úhlu, kde se zjišťovalo, zda se jedná o materiály hydrofilní nebo hydrofobní. Tyto ve vodě nerozpustné materiály byly dále biologicky testovány.
