Možnosti nanovláken jako nosiče pro preferenční růst specifických bakterií z aktivovaného kalu

Abstract
Tato práce se zaměřuje na hodnocení dvou nanovlákenných materiálů z polyamidu 6 a polyuretanu použitých jako nosiče pro nitrifikační bakterie. Vlákna byla připravována pomocí střídavého elektrického pole. Testovacím zařízením byl bioreaktor s fixním ložem, na kterém byla vlákna připevněna. V pravidelných týdenních intervalech byly odebírány vzorky jak pro otestování aktuálního stavu bioreaktoru, tak pro zhodnocení růstu a složení uchyceného biofilmu na nosičích (fluorescenční in-situ hybridizací, FISH). Optická mikroskopie sloužila k porovnání vizuálních aspektů zaplnění nanovlákenné plochy biofilmem. Správnou volbou nanovlákenného pokryvu (materiálu) byl podpořen růst nitrifikačních bakterií ve formě biofilmu. Správnost této volby potvrdilo výrazné snižování podílu všeobecných bakterií na vláknech PU ve prospěch většího zastoupení bakterií nitrifikačních.
This work focuses on the evaluation of two nanofibrous materials made of polyamide 6 and polyurethane used as carriers for nitrifying bacteria. Fibers were prepared using the alternating electric field. The test device was a fixed bed bioreactor on which the fibers were attached. Samples were taken at regular weekly intervals for both testing the bioreactor condition and assessing the growth and composition of biofilm captured on carriers (fluorescence in situ hybridization, FISH). Optical microscopy served to compare the visual aspects of filling the nanofibrous area with biofilm. By choosing the nanofibrous coating (material) we supported the growth of nitrifying bacteria in the form of biofilm. The accuracy of this choice has been confirmed by a significant reduction in the proportion of generic bacteria on PU fibers in favor of a higher proportion of nitrifying bacteria.
