Nanovlákenné kožní kryty pro použití ve veterinární medicíně

Abstract
Tradiční krytí, jako jsou obvazy a gázy, mohou narušovat hojení kožního traumatu při výměně. Zvlášť ve veterinární medicíně je vyžadováno rychlé a přesné ošetření, bez traumatizujícího převazování. Nabízí se vyrobit novou náplast, kterou není potřeba vyměňovat, protože se po čase sama vstřebá. Nanovlákna se přirozeně nacházejí v mimobuněčné hmotě, a proto je jejich použití příhodné. Tato práce se zabývá výrobou potencionálního kompozitního kožního krytu z nanovlákenných vrstev vyrobených bezjehlovým elektrostatickým zvlákňováním. Krycí vrstva je z biodegradabilního polyuretanu, který dočasně přebírá zejména ochrannou funkci kůže. Lepší adhezi k ráně zajišťuje hydrofilní polyvinylpyrrolidon. Jejich vhodným spojením může vzniknout kompozit, který zajišťuje nepřerušované hojení. Navíc byly do vrstev přidávány aditiva, jód za účelem dezinfekce a propolis pro podporující účinky. Bylo dosaženo cíle vytvoření kompozitního materiálu pomocí zvlákňování ze směsi polymerů a laminováním dílčích vrstev. Tento postup byl proveden také s pilotní koncentrací aditiv. Testování materiálů proběhlo pomocí skenovací elektronové mikroskopie, měření kontaktního úhlu a kvalitativní analýzou aditiv. Biologická kompatibilita byla testována pomocí stanovování cytotoxicity extraktů materiálů s buněčnými liniemi NIH3T3 a HaCaT a in vitro testy s vyhodnocením 1., 3., 7. a 14. den pomocí metabolického CCK-8 testu a fluorescenční mikroskopie.
Changing traditional wound dressings, such as bandages and gauzes, can interfere with the wound healing process. In veterinary medicine it is especially necessary to effect fast and precise treatment without the trauma of re-bandaging. It is therefore appropriate to consider the production of a new plaster which does not require changing, as it would be self-absorbing after a certain amount of time. Nanofibers are naturally occurring in extracellular matrices and therefore their use is appropriate. This thesis deals with the manufacture of a potential composite wound dressing of nanofibrous layers using needle-less electrospinning technology. The cover layer is biodegradable polyurethane, which temporarily takes over the protective function of the skin. Improved adhesion to the wound is provided by hydrophilic polyvinylpyrrolidone. In the appropriate combination, this creates a composite, enabling uninterrupted wound healing. Furthermore, the additives iodine (for disinfection) and propolis (to enhance healing) were added in layers. The objective of this thesis - to create a composite wound dressing - was achieved by blend electrospinning and laminating sectional layers. Materials testing was carried out by scanning electron microscopy, measuring the contact angle and the qualitative measuring of additives. Biocompatibility was tested by cytotoxicity of material extracts with cell lines NIH3T3 and HaCaT and in vitro testing with results from the 1st, 3rd, 7th and 14th day by metabolic CCK-8 assay and fluorescence microscopy.
