Nanovlákenná vrstva z PVDF a její modifikace pro potlačení fibrotické reakce

Abstract
Tato bakalářská práce se zabývá nanovlákennou vrstvou z polyvinylidenfluoridu a její modifikací pro potlačení fibrotické reakce. Fibrotická reakce je jednou z hlavních příčin pooperačních komplikací u onemocnění glaukomu, proto je důležité, aby tento problém nenastal, nebo byl alespoň minimalizován. Tomu by mělo zabránit antifibrotizační činidlo mitomycin C, který bude vlákenná vrstva z polyvinylidenfluoridu obsahovat už od samého začátku. V teoretické části je vysvětleno, z jakého důvodu byl vybrán tento polymer s konkrétními rozpouštědlovými systémy a jeho další využití ve tkáňovém inženýrství. Také budou blíže popsány jednotlivé části oka, které při poškození vyvolávají oční onemocnění glaukom, a to konkrétně: řasnaté tělísko, trabekulární síťovina a oční nerv. Dále bude vysvětlena problematika adheze buněk a fibróza tkáně, která je pro tuto bakalářskou práci klíčová. Experimentální část je zaměřena na optimalizaci polymerního roztoku a výrobu vlákenné vrstvy pomocí dvou způsobů elektrostatického zvlákňování, kdy je následně zkoumána jejich morfologie. Dále jsou materiály zkoumány pomocí in vitro testování, kdy je použito antifibrotizační činidlo, které by mělo potlačit fibrotickou reakci.
This bachelor thesis deals with polyvinylidenfluoride nanofibrous layer and its modification to prevent of fibrotic reaction. The fibriotic reaction is one of the main causes of postoperative complications in glaucoma therefore it is important to prevent or at least minimaze this index. This should be prevented by the antifibrotizing reagent mitomycin C, which will the fibrous layer of polyvinylidenfluoride contain from the beginning. Furthermore theoretical part explains why this polymer and its solvent systems were chosen and what are its other uses in tissue engineering. There are also more precisely explained functions of the individual parts of the eye that cause glaucoma especially the ciliary body, the trabecular meshwork and the optic nerve. There is also explained cell adhesion and tissue fibrosis which are important for this bachelor thesis. The experimental part is focused on the optimization of the solution and the production of a fibrous layer by two methods of electrostatic spinning. After that is examined their morphology. Furthermore, the materials are subjected to cytotoxicity tests, due to an antifibrotic reagent which should suppress the fibrotic response.
